Epicutaneous vaccination with protease inhibitor-treated papain prevents papain-induced Th2-mediated airway inflammation without inducing Th17 in mice

Environmental allergen sources such as house dust mites contain proteases, which are frequently allergens themselves. Inhalation with the exogenous a model of protease allergen, papain, to airways evokes release and activation IL-33, promotes innate adaptive allergic airway inflammation Th2 sensitization in mice. Here, we examine whether epicutaneous (e.c.) vaccination antigens without activity shows prophylactic effect on Th Th2-medated inflammation, driven by or endogenous IL-33. E.c. ovalbumin restrained ovalbumin-specific and/or subsequent inhalation plus papain recombinant inhibitor-treated papain-specific Th9 sensitization, eosinophilia, infiltration IL-33-responsive group 2 lymphoid cells papain. However, e.c. but not induced Th17 response bronchial draining lymph node cells. In conclusions, demonstrated that via intact skin mice even allergen-activated IL-33-driven attenuate attenuated similar its derivative lacking activity, although former latter promoted development. addition, present study suggests modified allergens, Th17-inducing adjuvant was eliminated, might be preferable for safer clinical applications administration.
